4.Does it loop seamlessly?

Not perfectly. The last frame is not matched to the first, so the restart is visible if you look for it. The motion is kept small and continuous so that restart is as unobtrusive as possible.

5.Can I get a transparent background?

No. The clip is opaque video and the GIF inherits that. For a cut-out subject, remove the background from the still image first and treat the result as a picture rather than a loop.
